In cellular respiration, anaerobic respiration (oxygen-free) occurs in the cytoplasm of the cell, whereas the majority of aerobic respiration (containing oxygen) occurs in the mitochondria.

Blood samples, aspirates of bodily fluids (pleural, pericardial, cerebrospinal, peritoneal, and joint fluids), urine obtained by suprapubic aspiration, abscess contents, deep wound aspirates, and specimens obtained through specialized procedures like transtracheal are among the materials suitable for anaerobic culture.

The following locations are unsuitable for anaerobic cultures due to resident anaerobic flora: sputum, stomach contents, throat and nasopharynx, bronchoscopy specimens, voiding or catheterized urine, urogenital swabs (such as vaginal and/or cervical), and specimens from superficial wounds.

• The nonselective medium for isolating facultative and anaerobes is anaerobic blood agar. 
• Egg-yolk agar (EYA): Nonselective for determining the amount of lipase and lecithinase produced by fusobacteria and clostridia.

Another technique that is unique to these bacteria is gas-liquid chromatography (GLC), which allows for the quick, accurate, and trustworthy identification of anaerobes based on the alcohols, organic acids, and short-chain fatty acids they create.
