What is an Acid Phosphatase Test?

Acid Phosphatase is defined as a lysosomal enzyme that hydrolyses the organic phosphates under acidic conditions. Although, the post-pubertal prostatic epithelial cell is seen to have a particularly high concentration of Acid Phosphatase level. This enzyme is also found in the bone, spleen, kidney, liver, intestine, and the blood.

Other Names:

AP, presumptive test for seminal fluid

Why is Acid Phosphatase Test done?

The acid phosphatase (AP) test is usually done to check for the presence of sperm in casework items. This color test is generally performed on filter paper, which is then kept after the test.

This is done to look for the following:

  1. Prostate Cancer/Injury/Infection
  2. Presence of Semen in the vaginal fluid after there has been alleged sexual assault on the victim and useful as an evidence in the court trial.

What is Forensic Acid Phosphatase?

Today, the Acid Phosphatase test is also being used by the forensic scientists, and considered an investigative tool.

It is seen that the Acid Phosphatase test. Forensics usually aids in the recognition of seminal stains in typical rape cases.

As Semen is known to contain Acid Phosphatase, this test is done to detect and evaluate the presence of the same.

The Significant acid phosphatase (ACP) and the prostate specific Acid Phosphatase (PAP) level will give a clear picture of the Rape Victims.

As this enzyme in present in abundance in the semen. Thus, the test can be used as a tool to notice the existence of acid Phosphatase substrate on the offenders' clothes, carpets, floors and vaginal and rectal parts of the victim.

This is then correlated with an intercourse that has taken place within the preceding 24 hours only.

Guidelines to follow before, during and after the test

Here are some guidelines an individual can refer to once they have decided to undergo Acid Phosphatase Test (Serum)

The required documents needed for this test includes your valid government ID and an appropriate Doctor’s prescription.

Before the test

  1. Nothing Specific to prepare for the Acid Phosphatase Test (Serum) in advance.

During the Test

Method: When blood is drawn, it is called “venipuncture”

  1. At the test center, you would be requested to sit comfortably on the chair or even lie down on the cot.
  2. In case of long sleeve clothing, roll up sleeve of one end or remove the arm from the sleeve of the clothing.
  3. A healthcare provider, or a technician/nurse, would tie band made of rubber tightly on your upper arm.
  4. Once an appropriate vein is found, a needle is inserted below the skin to take the sample of about 2ml.

A short sharp prick may be felt when needle puncture is made under the skin, to collect the blood sample.

After the test

  1. After the blood is withdrawn, the respective technician/ healthcare professional would withdraw the needle.
  2. Place pressure on puncture wound to stop the bleeding. A small bandage may also be put on the wound.
  3. You would be able to return on your usual routine immediately.

For Acid Phosphatase, Prostatic Test

How does one prepare for the Test?

Please avoid the drugs given below for at least the next 24 hours before the sample is collected:

  1. Androgens in females.
  2. Clofibrate Drugs.
  3. Antilipidemic drugs.

Note: It is important to NOT STOP TAKING ANY MEDICINE without consulting the respective healthcare professional.

How to Interpret the Results?

Result Interpretation

To interpret results, it is important to understand the Acid Phosphatase normal/ desired range in the body is equal to the value of 2 ng/mL.

The Prostatic Acid Phosphatase, the normal range is used as the base which will be used to interpret the results, from the undertaken PAP test.

According to the healthcare professionals, the normal PSA evaluated in blood is around 4.0 ng/mL.

A value higher than this signifies present Prostrate Cancer.

FAQs on Acid Phosphatase Test

What is serum Acid Phosphatase?

The Acid Phosphatase is the enzymes which are capable of hydrolyzing the phosphate esters in an acidic environment. The Prostatic Acid Phosphatase (PAP), is produced in prostate of the men, it is considered to be a major serum marker for detection of prostate cancer.

What is the Acid Phosphatase test also known as?

It is known as: Serum Acid Phosphatase test. This also evaluates the level of PSA in the blood. In an Acid Phosphatase assay kit, it could be used to detect the level of the enzymes present in the blood serum.

Why is Acid Phosphatase test important?

The Prostate Acid Phosphatase (PAP) is known to be used extensively as the serum marker for diagnosing cancer of the prostate gland. It is also later released in the serum. The prostate gland is seen as to be in increasing amounts in a malignant tissue as it keeps proliferating.

What is the principle of Acid Phosphatase test?

The enzyme Acid Phosphatase is seen hydrolyzing the substrate to phenol and the phosphate ion. This phenol formed is then also simultaneously coupled with the suitable diazonium salt, called chromogen. This gives a characteristic colored, which then is detected as a positive test for seminal stain presence.

What is the diagnosis of Acid Phosphatase?

ACP test is used normally used to diagnose and also the assess conditions such as the cancer of the prostate (this helps in diagnosing and monitoring the effect of treatment).

What disease is related to Acid Phosphatase?

It is also seen that in some bone diseases such as (Paget's disease) or the Prostatitis (i.e. infection or inflammation of the gland), the hyperparathyroidism, benign hypertrophy of prostate gland (BPH), Osteopetrosis etc.

What is the normal value of acid and alkaline Phosphatase?

Normal Values:

The Total Acid Phosphatase in the serum: 5.5 U/l. (25 °C); in prostatic Phosphatase: up to 2 U/l. (25 °C). Also, the level of alkaline Phosphatase in serum is usually: 18–63 U/l.

What does high Acid Phosphatase mean?

Acid Phosphatase: This enzyme liberates the phosphate under the acidic conditions and is usually produced in the liver, the  spleen, the the bone marrow, and the prostate gland.

Abnormally high serum level: Indicates an infection, an injury, or the cancer of the prostate.

What is the significance of Acid Phosphatase determination?

Clinical Significance
In around 60% of men with the prostatic cancer in cases of metastases, the value is seen to be high.

Acid Phosphatase Test for Female

It has been seen that the  even though the Serum Acid Phosphatase activity is normal, the OC-derived tartrate-resistant Acid Phosphatase has been  inexplicably elevated for more than a decade in affected women. There also have been an  Increased level of proline in blood and  the urine that have been described in few patients, however, the significance has not been known.

What is the Acid Phosphatase test for leukemia? Leukocyte Acid Phosphatase

This test generally looks at chemical tartrate-resistant acid phosphatase (TRAP) in the blood. The test can be carried out on the bone marrow. The TRAP can thus appear in the blood in case there is an injury and have hairy cell leukemia.

What color is produced with a positive Acid Phosphatase test?

Positive Result:  A Purple color will be seen within 3 min itself (Acid Phosphatase Color test).
