What is IDH-1 test?
IDH stands for Isocitrate dehydrogenase. It is an enzyme that act as a catalyst in the oxidative decarboxylation of isocitrate leading to the formation of alpha-ketoglutarate and carbon dioxide (CO2).
Isocitrate is a component of citric acid cycle, also known as Krebs cycle. The reaction catalyzed by the IDH enzyme is usually irreversible and therefore it must be carefully regulated in order to:
- Prevent depletion of Isocitrate
- Avoid accumulation of alpha-ketoglutarate.
There are 3 isoforms of IDH in humans namely:
- IDH 1
- IDH 2
- IDH 3

- After processing, DNA is extracted from the samples.
- PCR is done to amplify DNA fragments containing IDH 1 and IDH-2 genes.
- Later gene sequencing is done.
- Reports are analyzed for IDH-1 and IDH-2 mutations.
