What is Diphtheria Culture Test?

The Diphtheria Culture test is used to detect the presence of Corynebacterium diphtheria bacteria. When diphtheria is suspected, this test is performed. Diphtheria is a potentially fatal bacterial infection of the respiratory tract. These bacteria can be found in the nose, throat, and skin. Clinical specimens for culture are collected from the nose or nasopharynx, as well as the throat, of all suspected cases and their close contacts. Swabs from beneath the membrane are also taken, or a piece of the membrane is removed. When diphtheria is suspected, specimens for culture are obtained.

Why it is done?

This test allows doctors to determine whether the bacteria C diphtheriae, which causes diphtheria, is present. When diphtheria bacilli are isolated, the presence of the toxin-producing gene is checked. A variety of in vitro and in vivo tests are used to determine the toxigenicity of C diphtheriae.

What is the procedure for Diphtheria Culture Test?

The patient is asked to open his or her mouth and tilt his or her head back. The doctor will use a tongue depressor to press the tongue down and examine the mouth and throat.

How does one prepare for the test?

Before undergoing the test, patients should avoid using any antiseptic mouthwash. Patients must notify their doctor if they are taking any antibiotics, as this may interfere with the tests. The patient should not gargle prior to the throat culture. Before the procedure begins, the patient's immunisation history is also checked.

FAQ

The bacterium produces a toxin that accumulates in your throat, making it difficult for you to breathe and swallow. A person suffering with diphtheria in a warmer area may also have non-healing skin lesions that may become coated in gray tissue.

It has been observed that the diphtheria bacterium can reside in the infected person's mouth, throat, and nose and can be spread to other people through coughing or sneezing. On rare occasions, skin sores or objects soiled by pus seeping from sick people's sores can spread the infection.

The confirmation of toxigenic C diphtheriae in throat or lesion cultures by bacteriologic laboratory testing is necessary for the clinical diagnosis of diphtheria. Different types of media, such as Mueller-Miller tellurite agar, Tinsdale tellurite agar, or Loeffler agar, can be used for initial isolation.

The Pertussis and Diphtheria Laboratory (PDL) accepts swabs from the nose, throat, or wounds for the purpose of cultivating C. ulcerans and C. diphtheriae cultures. Use swabs made of nylon, polyester, or rayon.

Blood agar is a good medium for the growth of diphtheria; however, tellurite blood agar, also known as Hoyle's medium, is advised since it inhibits other respiratory flora and promotes the development of the three biotypes' distinctive colonial morphology (gravis, intermedius, and mitis).

Diphtheria in close proximity, in addition to a positive patient PCR test, can bolster the Corynebacterium diphtheria diagnosis.  When cultured in Loffler's media and stained with polychrome methylene blue (Albert stain), the result is a metachromatic stain.

The level of diphtheria antibodies in your blood that result from receiving a diphtheria vaccination is measured by this test. These might potentially result from a prior diphtheria infection.
