What is Proliferating cell nuclear antigen (PCNA) Test?

PCNA, Proliferating Cell Nuclear Antigen Nuclear Antigen is a protein found only in cancerous and proliferating cells. This is a nuclear antigen that is involved in DNA synthesis, repair, and cell cycle progression. During DNA replication, PCNA is required for the coordinated synthesis of both the leading and lagging strands at the replication fork. PCNA expression is linked to the proliferation of both malignant and cell types. Patients with systemic lupus erythematosus have these antibody PCNA cells (SLE). This Anti PCNA test is performed on a sample of blood drawn from the patient in order to quantify the levels of Anti PCNA in the blood and to confirm SLE, as well as during and after treatment for systemic lupus erythematosus.

Other Names:

Anti PCNA florescent Techniques

Why it is done?

Anti-PCNA antibodies are well known for detecting activated lymphocytes and malignant proliferating cells, making them an excellent clinical marker for systemic lupus erythematosus (SLE). PCNA's unique structure allows it to boost the processivity of DNA polymerase delta. It plays an important regulatory role in the initiation of leading strand DNA replication and the termination of lagging strand synthesis. PCNA can collaborate with a wide range of cell cycle proteins and helps to manage a number of proteins involved in DNA processes such as apoptosis.

What is the procedure for Proliferating cell nuclear antigen (PCNA) Test?

Anti-PCNA antibodies can be detected using Immunofluorescence (IF), Double Immunodiffusion (DID), Counter Immun electrophoresis (CIE), Enzyme-Linked Immune Sorbent Assay (ELISA), and Immunoblotting (IB) methods on a patient's blood sample. The ELISA method is commonly used because it is reliable and produces accurate test results. The ELISA kit is used for the test. The collected blood sample is tested on the solid surface of the kit, which usually has an antibody attached to it. The results of the tests are forwarded to your doctor for further analysis and treatment.

How does one prepare for the test?

There isn't much preparation required. Your blood sample is collected by the lab technician for the test. Only if you're taking any medications, have allergies, or have underlying medical conditions should you inform your doctor, and the doctor will advise you on any precautions to take before, during, or after the test.
